KSM-66 Ashwagandha: Why the Extract Matters More Than the Plant

Withania somnifera — commonly known as ashwagandha — is an adaptogenic herb used in Ayurvedic medicine for centuries. In modern nutritional science, it is among the most studied botanical adaptogens, with a growing body of randomized controlled trials examining its effects on stress markers, cortisol, testosterone, and cognitive function.
The active compounds are primarily withanolides — a class of steroidal lactones that give ashwagandha its pharmacological properties. The concentration of withanolides varies significantly between raw root powder, whole root extracts, and standardized extracts.
What standardization means
A standardized extract is one in which the manufacturing process is controlled to ensure a consistent concentration of active compounds in every batch. KSM-66, developed by Ixoreal Biomed, is a full-spectrum ashwagandha root extract standardized to a minimum of 5% withanolides.
A raw ashwagandha root powder may contain anywhere from 0.5% to 3% withanolides depending on the plant source, growing conditions, and processing. A non-standardized extract may perform inconsistently between batches.
When a clinical study reports benefits from ashwagandha at 300mg or 600mg, that study almost always used KSM-66 or a comparably standardized extract — not raw powder. Assuming equivalent effect from raw powder is a category error.
What the research shows
A double-blind, randomized, placebo-controlled study published in the Indian Journal of Psychological Medicine (Chandrasekhar et al., 2012) found that participants taking 300mg of KSM-66 twice daily for 60 days showed significantly reduced scores on perceived stress scales and significantly lower serum cortisol levels compared to placebo.
A study in the Journal of the International Society of Sports Nutrition (Wankhede et al., 2015) found that KSM-66 supplementation at 300mg twice daily was associated with significant improvements in muscle recovery, strength, and testosterone levels in resistance-trained men over 8 weeks.
A study in Medicine (Choudhary et al., 2017) found improvements in memory, executive function, and sustained attention with KSM-66 supplementation at 300mg twice daily over 8 weeks in adults with mild cognitive impairment.
What this means for your supplement
Check the label. If it says "Ashwagandha root powder" without specifying a standardized extract and a withanolide percentage, you cannot know what you are getting. If it says "KSM-66 Ashwagandha Extract (5% withanolides)," you are getting the form the research was done on.
